What Retatrutide Was Supposed to Do, MOTS-c Might Actually Do Better
One of the most fascinating potential fat-loss combinations isn’t retatrutide.
It’s tirzepatide + MOTS-c.
And the science behind MOTS-c is incredibly interesting.
MOTS-c is a naturally occurring, 16-amino-acid peptide encoded by your mitochondrial DNA.
Your mitochondria aren’t just responsible for producing energy. They also communicate with the rest of your cells to regulate metabolism, stress adaptation, and cellular health.
MOTS-c is part of that communication system.
Here’s where things get exciting.
MOTS-c activates AMPK, one of the body’s key cellular energy sensors.
AMPK helps your cells respond to energy demand by increasing glucose uptake, promoting fatty acid oxidation, and improving metabolic flexibility.
Essentially, it helps regulate how efficiently your body uses carbohydrates and fats for energy.
In a landmark 2015 study, MOTS-c prevented diet-induced obesity and insulin resistance in mice.
Researchers discovered that MOTS-c influences the folate-purine pathway, increasing AICAR, a molecule that activates AMPK.
But here’s what really caught my attention.
MOTS-c may also help preserve muscle.
Myostatin is a protein that acts as a natural brake on muscle growth.
In a 2021 study, researchers found that MOTS-c reduced myostatin expression and protected against muscle atrophy in experimental models.
It also activated the AKT pathway, which helps regulate muscle protein metabolism, and inhibited FOXO1 activity, a transcription factor involved in activating muscle-wasting genes.
Think about the implications.
During aggressive dieting, the challenge isn’t simply losing weight.
It’s encouraging fat loss while minimizing muscle loss.
And MOTS-c appears to influence pathways on both sides of that equation: improving energy metabolism while potentially reducing signals associated with muscle breakdown.
There’s another fascinating connection.
Exercise naturally increases MOTS-c.
Human research has demonstrated that exercise increases MOTS-c expression in skeletal muscle and circulation.
And in animal studies, administering MOTS-c improved exercise performance, even in older mice.
This suggests MOTS-c may be involved in some of the metabolic adaptations associated with exercise.
Now consider combining this with tirzepatide.
Tirzepatide activates GLP-1 and GIP receptors, improving satiety, reducing food noise, and helping regulate glucose metabolism.
MOTS-c potentially complements this through AMPK activation, improved metabolic flexibility, and muscle-protective signaling.
That’s a fascinating combination from a body-composition perspective.
Retatrutide takes a different approach by adding glucagon receptor activation, increasing energy expenditure and amino acid breakdown.
My interest in MOTS-c is whether we could improve fat utilization while simultaneously supporting muscle preservation.
Of course, there’s a major caveat.
MOTS-c is still experimental.
Most of this research comes from cells and animals. We don’t yet have human clinical trials demonstrating that administering MOTS-c preserves muscle during dieting, produces meaningful fat loss.
I haven’t personally used MOTS-c.
But it’s a peptide I’m increasingly interested in researching.
And MOTS-c has some genuinely fascinating biology worth investigating.
Will be adding bpc 157 and tb500 to kino clinic next week
I’ve used intermittently for injuries that were stuck and no longer healing.
It’s been helpful. But it takes a solid 6-8 weeks.
Will be looking at adding MOTS-c as well
In poor metabolic states like obesity and type 2 diabetes, glucagon activity is often elevated.
Tirzepatide has been shown to suppress glucagon secretion, improving metabolic regulation.
Glucagon also plays a role in amino acid breakdown in the liver, making lowering this pathway particularly interesting for muscle preservation.
Tirzepatide activates two receptors: GLP-1 and GIP.
This improves satiety, reduces food noise, and supports better glucose regulation and energy utilization.
We’re also seeing broad metabolic benefits: improved insulin sensitivity, healthier lipid profiles, and reductions in markers of inflammation.
This is one reason I personally use 2-3 mg of tirzepatide per week.
It’s made staying lean remarkably seamless, without feeling like I’m constantly dieting.

THE GREAT RETA LIE
What if retatrutide’s third receptor is actually a disadvantage for people who want to lose fat and preserve muscle?
Everyone is celebrating that retatrutide activates THREE receptors instead of two.
But that third receptor is glucagon.
A catabolic hormone.
Glucagon signaling can increase energy expenditure, potentially allowing you to burn more calories per day (to the tune of 70-250).
Not bad.
But HOW does it increase energy expenditure?
One of glucagon’s well-established actions is increasing amino acid uptake and breakdown in the liver.
Essentially, you’re increasing amino acid catabolism.
Why in God’s green earth would you WANT to increase amino acid breakdown when your entire goal is to lose fat and preserve muscle?
“But I’ll just eat more protein!”
Okay. But now you’re eating more calories to compensate for the increased amino acid oxidation.
You’re offsetting the additional calories you were trying to burn.
Wouldn’t it be better to avoid unnecessarily increasing amino acid catabolism in the first place?
And that’s before considering retatrutide significantly increases resting heart rate.
But what does the actual body composition data say?
In separate phase 2/body composition analyses, approximately 37% of weight lost with retatrutide has been cited as lean mass, compared with roughly 25% in the tirzepatide SURMOUNT-1 substudy.
That’s a substantial difference.
It raises an important question: Is most of retatrutide’s additional weight loss coming at the expense of lean tissue?
I think so.
This is why I’ve favored tirzepatide for my fat-loss and body-composition goals and for my clients.
And why I personally use 2.5 mg of tirzepatide per week.

Problem with x is it has the worst video algorithm of any social media platform
It is absolute trash
Many people I know just deleted it
I delete is all the time cuz of how terrible the video algorithm is
It’s just horrible things
All negativity
I will forever love tirzepatide for allowing me to eat plenty of spicy rigatoni while staying lean
This was something I could never accomplish before
I’d just eat and want more and more lol
Reta works very well
But I prefer tirzepatide
It’s cleaner.
You’re not hitting a catabolic hormone and raising up heart rate significantly
Better muscle retention on tirzepatide shown in the data
More safety data and less side effects on tirzepatide
Reta produces slightly more weight loss but tirzepatide wins overall
And many have now switched over to tirzepatide and prefer it
You get better control over your appetite with tirzepatide
With Reta higher doses create faster heart rate. Not ideal
Be warm, positive and unaffected
These guys that get into arguments with women and try to put them in their place and get the last word
Is the gayest shit I’ve seen
Life is a two-way mirror. The world reflects the energy you bring into it.
If you walk into every interaction expecting hostility, rejection, and competition, that’s exactly what you’ll keep finding.
You’re making fundamental mistakes in how you perceive and interact with reality, then blaming the world for reflecting them back to you.
Choose the reality you want to experience. Hold that image. Give absolutely zero energy or reaction to anything that doesn’t align with it.
Stop fighting life. Flow with its currents.
The world isn’t against you. You’re against the world.
I was quickly reminded why I hate going out last night
Everybody is just so fucking mean and angry man. Always looking for a way to socially win against you. Girls will bait you into a convo to walk away. Guys will find a way to get a quick “nut drag” on you socially
People will find ways to just throw social jabs when they are supposed to be out having a good time
I’m starting to genuinely hate the human race
I used to think our one redeemable quality is our thinking and rational abilities to transcend us from a beast
This genuinely doesn’t exist in most people
Full body workouts are overrated and unenjoyable
Hitting a muscle 3x per week is silly because there’s no way you can hit 3 PR’s per week on the same muscle
You are often training a muscle when it’s not even fully recovered
Upper and lower splits not ideal tbh
Hitting legs twice per week not great for lifestyle. You end up just hungry and sore and legs take a lot of the gains not upper
Just hit
Chest, biceps, rear delts
Lowerbody
Shoulders, back, triceps
And focus on hitting PR’s each week
Muscle growth is directly tied to strength gain on key lifts
Many top bodybuilders like mentzer and Yates just trained 3x per week and focused on strength gains on key movements not chasing high volume or high frequency
Natural sway on lat pull downs will build more muscle than super strict vertical form
You will hit more personal records
And you will engage your traps and rhomboids for more overall growth
Just keep the form consistent
But will you have to be on a glp-1 for life?
I don’t understand this question
If you like being on it, stay on it ???
If you don’t, go off of it ???
There’s a ton of hype around GHK-Cu right now, both topical and injectable.
But the evidence is still very limited.
Meanwhile, most people are overlooking one of the most powerful, well-researched anti-aging topicals we have: tretinoin.
Tretinoin is a prescription-strength vitamin A derivative with decades of clinical research behind it.
It increases skin cell turnover, stimulates collagen production, improves skin texture, reduces fine lines, and helps reverse signs of sun damage.
Over time, it can even increase the thickness of the skin’s deeper layers.
The catch? It takes consistency. You’ll typically need 3–6 months to see meaningful improvements, with benefits continuing beyond that.
You also need to introduce it gradually, manage irritation, and be diligent about sun protection.
It’s not some exciting new peptide. It’s a proven treatment that’s been around for decades.
The best tools aren’t the newest or trendiest. They’re the ones with the strongest evidence.
The irony of Bryan Johnson’s obsessive pursuit of longevity is that he’s taken the same mindset that burned him out in business and applied it to anti-aging.
Endless optimization. Obsessive tracking. Sacrificing the present for some future outcome.
He learned the wrong lesson.
He thought the lesson was to stop sacrificing his health for money.
But the deeper lesson was to stop sacrificing the present moment for some imagined future.
To stop treating life as a means to an end. A stepping stone to something better. An obstacle to overcome.
Damnnn you better looks max to get a girl lol
Cuz she won’t need you to buy her stuff
🚨 Elon Musk has dropped an unprecedented bombshell—right inside the White House, in front of Donald Trump: 💥🇺🇸
“From now on, you will no longer need to work for any company... I will make sure everyone on Earth enjoys a celebrity-level lifestyle and income!”
Musk is completely challenging the traditional concept of government-provided “Universal Basic Income” (UBI) designed merely for survival, presenting a radical vision of a new form of capitalism.
According to Musk, how will everyone become wealthy? 👇
As AI and robots produce goods and services 24/7 without limits, the overall standard of living for all humanity will skyrocket—as if everyone were earning an astronomical salary.
Musk confidently declares: “Everyone will have access to better healthcare than I personally receive!” Through the combination of superintelligent AI and precision surgical robots, top-tier medical care could become available to everyone at virtually no cost.
AI will handle the thinking, while robots build roads and structures, grow crops, and manufacture products. With forced labor becoming a thing of the past, the phrase “I don't have enough money to buy what I need” could disappear from the human vocabulary entirely!
While 99% of humanity worries that “AI will take their jobs,” Musk envisions a future where an army of autonomous robots will turn every single person into a king—living a life of total ease!
