Just a periodic reminder: RNA abundance ≠ transcription activity. RNA abundance is also regulated by RNA degradation.

We were pretty stoked to be the first investor in @KernaLabs They've been heads down building and have just published a detailed essay on the future of RNA medicine. kernalabs.substack.com/p/mrn… More to come.

Melissa built one of the first mRNA platforms at Moderna. Now she's writing about the future of mRNA in the era of AI
AI is rapidly expanding the supply of therapeutic ideas, but turning them into effective medicines is still hard. In our new essay, our co-founder Melissa J. Moore explores how better mRNAs can unlock novel medicines, and why mRNA’s most important chapter is still ahead. kernalabs.substack.com/p/mrn…

Programmable medicine has the potential to solve some of our most pernicious chronic diseases. I’m profoundly excited by the future that @KernaLabs is bringing to life. With their work, I’m optimistic that many of the diseases that erode our quality of life can soon be eradicated.

Moderna just showed what mRNA can do beyond vaccines: its individualized cancer therapy, with Keytruda, succeeded in Ph3. When LLMs began rapidly progressing 3y ago, we made a bet that AI models can outperform human-led design to create better mRNAs. 🧵1/4

Despite its obvious misuse during Covid, mRNA has tremendous promise for curing diseases. Artificial RNA essentially makes curing diseases a software problem.

We started Kerna on the thesis that mRNA would become a widespread therapeutic modality beyond vaccines. Today, Moderna’s mRNA cancer therapy + Keytruda met its Ph3 endpoints. We’re already building the foundation models and in vivo data engine for what comes next.

We're growing our investment team at Gradient! If you're looking to invest with the best in seed-stage AI, apply through the link below or ping me directly. 1/ This is one of the most unique times to invest at the seed stage: Capital is everywhere and software has been mostly commoditized.

Stumbled upon a weird LLM failure mode: if you ask Sonnet 4.6 to translate a DNA sequence to protein, the output includes Cyrillic characters ~50% of the time. The frequency is sensitive to spacing, which suggests a tokenization issue. Short writeup: erle.bio/blog/2026-03-22-fai…
