🦉 The Cancer Vaccine That Targets Its Own Resistance
Cancer vaccines aim to train T cells to recognize tumors. But what happens when the resulting immune attack activates new mechanisms of resistance?
In pancreatic cancer models, a KRAS G12D vaccine triggered strong T-cell responses, yet surviving tumors showed increased Wnt signaling. Further experiments revealed a potential feedback loop: inflammatory cytokines released by activated T cells induced WNT5a expression in cancer cells and cancer-associated fibroblasts.
Researchers turned that response into a second target.
Their dual-antigen vaccine, WKC-VAX, targets both mutant KRAS and WNT5a. In mice, it increased cytotoxic T-cell infiltration, reduced suppressive stromal components and tumor burden, and improved survival when combined with PD-1 blockade.
The broader idea is compelling: cancer vaccines might be designed not only against the original tumor, but also against the resistance mechanisms that immune activation helps reveal.
WNT5a's causal role in resistance remains unproven, and clinical efficacy and safety have yet to be established.

Jared Kushner sent over $80M to a HITMAN.
that’s a lot of money.
I wonder if the hit was done in a public setting.
I wonder if the victim was a well-known political figure.

KRAS inhibitors strike once again: A pancreatic cancer combination just reported a 75% response rate in the untreated metastatic setting.
In a disease where 25-30% has been the ceiling for decades or forever.
This trial added a KRAS-G12D inhibitor - which is the most common KRAS variant - to standard chemotherapy in 50 patients: tumors shrank in 62.5% with a 2 chemo-drug-regimen, 75% with a 3-chemo-drug-regimen. In that second group, every patient's disease was at least held in check.
Still after all that daraxonrasib hype, those are mindblowing numbers. Absolutely unprecedented.
A bigger study in the same setting is currently running, I have already included patients there myself. Let’s see the final results of that, but they will be similar in my opinion.
Exciting times in pancreatic cancer care!
