Manuscript is now published in Genomics. Conclusions are unchanged but text and figures have been improved after peer review. A big thank you to my collaborators that made the study possible. doi.org/10.1016/j.ygeno.2024…
Happy to present our new preprint! We ran a head-to-head comparison of sample multiplexing reagents for single-cell RNA-Seq on the same samples on the same day. 1/9 biorxiv.org/content/10.1101/…

Can’t wait to share more pretty images and awesome data revealing regulators of intrahepatic bile duct morphogenesis during my poster session at the @APSPhysiology Summit this Friday 5:15-6:05 (poster # P0753).
It was a difficult decision, but the results are in from our Pin It to Win It image contest!
Congrats to @HannahHrncir & her beautiful #microscopy of bile ducts!
#microscopymonday #liver #sciart @graczlab

ALT Hannah Hrncir Mouse left liver lobe intrahepatic bile ducts labeled with EpCAM and imaged in 3D with light sheet microscopy. The hierarchical nature of intrahepatic bile ducts with large ducts at the liver hilum that get smaller as they move towards the periphery is clearly illustrated. 'The tree of life'. Proper IHBD structure is essential to life.

So, essentially the problem is that replicates are needed to estimate biological/technical error. Once you have this, “bulk” methods are very good. However, with commonly used “droplet” methods, replicates are expensive and prone to batch effects. But SPLiT-seq is multiplexable.
