Congrats again to Yang Gao and the entire team for their pioneering research! Stay tuned for more of our work on this exciting target.
Interestingly, using the biochemical assays developed in this publication, we tested ZYS-1 (which I noted last week is not a genuine ADAR1 inhibitor) and confirmed it fails to inhibit ADAR1 even at concentrations as high as 1 mM.
Yang is a truly exceptional structural biologist, and I'm privileged to collaborate with his team on this target. Building on these mechanistic insights, we're making excellent progress developing ADAR1-targeted therapeutics.
This research elegantly demonstrates that ADAR1's activity depends not only on sequence but also on RNA-duplex length and structure. These findings are particularly significant given the enzyme's critical role in regulating innate immune responses.
Their recent Molecular Cell publication, "Biochemical Profiling and Structural Basis of ADAR1-mediated RNA Editing," provides atomic-resolution insights into how ADAR1 interacts with endogenous RNA substrates.
Congratulations to Xiangyu Deng and Yang Gao at @RiceUniversity for their outstanding work unraveling the complexities of RNA editing! Biochemical profiling and structural basis of ADAR1-mediated RNA editing: Molecular Cell cell.com/molecular-cell/full…
This underscores the need for target-specific assay validation in drug discovery. Join the conversation and read our full preprint for a detailed discussion. #ADAR1 #DrugDiscovery #RNAEditing #CancerResearch #AssayValidation
Our results revealed that ZYS-1 does not inhibit ADAR1 activity at concentrations up to 1 mM, challenging previous conclusions based solely on binding data.
In our study, we employed a rigorously validated ADAR1-specific biochemical assay that directly quantifies inosine formation on physiological dsRNA substrates.
Wang et al. used an ADA assay, which is designed to measure free inosine, not inosine incorporated within dsRNA. This calls into question the assay’s suitability for ADAR1 activity.
ADAR1 is a key RNA-editing enzyme that converts adenosine to inosine in dsRNA—a process essential for modulating innate immunity and a highly pursued target in cancer therapy.
Excited to share our new preprint on @biorxivpreprint , where we challenge the claim that ZYS-1 is a potent ADAR1 inhibitor, as reported by Wang et al. in @NatureCancer. Re: Concerns Regarding the Validation of ZYS-1 as a Bona Fide ADAR1 Inhibitor biorxiv.org/content/10.1101/…

These cuts violate the law, hurt people, and undermine the very system of scientific research & groundbreaking advancements that we are so proud happen in #TX07.
