LRK-4189: A Potential First-in-Class PIP4K2C Degrader Heads to the Clinic | drughunters.com/474hvrI PIP4K2C has minimal catalytic activity, but reported scaffolding roles make it well-suited for targeted degradation. Building on insights from tool inhibitors and an academic degrader, Larkspur optimized a CRBN-recruiting molecule, LRK-4189, now planned for first-in-human study in MSS colorectal cancer. Ex vivo CRC spheroid responses, supportive once-daily PK/PD, and xenograft proof-of-concept underpin the program. Read it on Drug Hunter: drughunters.com/474hvrI

Thanks to @MaverickNY of Biotech Strategy Blog for her insightful take on why PIP4K2C stands out as a cancer fitness target and how LRK-4189, our 1st-in-class degrader of this lipid kinase, has the potential to treat MSS CRC. #Biotech #Cancer
Time to talk turkey on how protein degradation may be a useful strategy for tackling a dark lipid kinase & overcome acquired resistance in CRC. Nice story from @LarkspurBio tinyurl.com/ynftnd7y
