Now online in @NatureSMB - We combine AlphaFold, structural, and biochemical approaches to show that SMO directly inhibits PKA via a pseudosubstrate mechanism—revealing a new mode of #GPCR signal transduction in the #Hedgehog pathway. Big thanks to everyone who contributed!

The new issue of #ScienceSignaling has arrived! Research in mice shows how microglia can trigger depressive-like behaviors linked to neuroinflammation, in vitro work reveals that adhesion GPCRs aren’t activated by corticosteroids, and more. scim.ag/4tLz8WI

A mechanistic analysis of adhesion #GPCRs reveals that peptide mimics, but not the corticosteroid beclomethasone, can activate signaling downstream of the GPCR GPR97/ADGRG3 and trigger neutrophil shape changes and migration. @UMPharmacology @lab_tall scim.ag/4kNU77g

Excited to share our latest preprint! We used cryoEM and MD simulations to look at the GTP-induced activation of the mu-opioid receptor-Gi complex with partial, full, & super agonists resolving Gi activation and how its modulated by different ligands! biorxiv.org/content/10.1101/…

Excited to present our preprint on the structure-function relationship of anaphylatoxin complement receptors. A casual number of 19 structures deciphering ligand recognition mechanisms, G protein/arrestin and species specific signaling. Check it out! biorxiv.org/content/biorxiv/…

Our GzESTY paper is online. We optimized a cell based assay to sensitively detect GPCR activation in response to ligand application or in complex mixtures containing endogenous ligands. GzESTY was designed to aid in GPCR deorphanization efforts. nature.com/articles/s41467-0… – at Brighton, NY

Maybe the most amazing microscope paper of the year just dropped: Live 4pi-SIM (a.k.a I5S*): nature.com/articles/s41592-0… Having worked with Mats Gustafsson, I can attest that this is both extremely hard, but also extremely cool! Kudos to the authors! *cell.com/AJHG/fulltext/S0006…

Introducing a new class of potent opioid analgesics that remain in the periphery (hence no respiratory depression) and can be locally activated with light (hence no constipation). As efficacy switches, they also provide insight into how opioids can switch from weak to strong agonists! Great collab with Seva Katritch, Cornelius Gati, and Josh Levitz. biorxiv.org/content/10.1101/…

How proximal is proximity labeling❓ Using DNA nanorulers we found that TurboID/BioID are largely contact labeling, while APEX2 also bears a major contact labeling pathway in parallel with diffusive labeling. biorxiv.org/content/10.1101/… Special thanks to Alice Ting for discussions!

1/6 🧵 I'm building what might be the smallest #opensource #spectrometer with an easy to use UI! I call it a pocket spectrometer. Combining #M5StickC(~$20) with #AS7341 sensor (~$16). My goal? A complete spectrometer that is barely larger than a sample cuvette itself.

I would appreciate community guidance on building out research computing resources from scratch. Rather than building/maintaining compute clusters, what are the best cloud-based, university-level research computing options? I have my experiences, but I want to hear from others.
