Low-kV but high-res
Building on the work of Richard and Chris, here is our latest work showcasing the potential of the 100 keV #Tundra microscope with the Ceta F and Falcon C. We achieved high-resolution structures for a variety of protein complexes🧵.🔬#100keV #CryoEM. Movie by @lingboyu

#cryoEM #MicroED with electron counting. Protein structure at 0.8A resolution. Ab initio phasing for two proteins facilitated by electron counting on a falcon 4 direct electron detector. A section of the map is shown below biorxiv.org/content/10.1101/…

If the time slot on Nov 18th didn't work for you, the recording is now available on Tundra. thermofisher.com/us/en/home/… I had a lot of fun creating this video clip, learning on the job😊
Join us to take a close look at Tundra and the beautiful ❤️ structures Tundra produces. The event is on Nov 18, register now: ter.li/y43xvf

Jiri was in our Tundra lab end of August this year. In this clip, you can have a sneak preview of our Tundra.

@DimpleKaria3 and @lingboyu will show some very high resolution structures from #Tundra and several screening cases of challenging proteins #CryoEM at 100kV. Join on 18th Nov to see more.

Totally agree. Shot #1 and #2 look like purely for the number. Without touching the carbon support, these would normally suffer greatly from charging and do not contribute to the high resolution, unless there is conductive support over the hole, such as carbon.

This is impressive but the most important thing is to establish the best strategy to get the highest resolution (not the highest number of particles or movies) within a given scope time.

#CAFR, 2.6Å map of Apoferritin, 6hrs of data with Ceta-F from Tundra (100kV cryo-TEM)
Another 1.2Å map of Apoferritin #CAFR, now from just 6hrs of data with new Falcon 4i detector. 2 exposures per/hole and data processed with #Relion4 alpha
Preprint👇 updated with SARS-CoV-2 3a ion channel, 62KDa membrane protein at 2.1Å! Another wonderful work by @SteveBrohawn group @kerndm and @chrismhoel pushing #CryoEM
SARS-CoV-2 3a preprint update, 2.1Å! map with @AbhayKot @thermofisher, processed by me and @chrismhoel. We now have new PL uptake assays and RuR as an inhibitor from @sonalimali6 (Bautista Lab) and new 3a mutants patched by Ben Sorum. Led by @SteveBrohawn biorxiv.org/content/10.1101/…

The pixel size used is 0.73A, not ridiculously high, I think. 1) Ceta at 100kV is way better than Ceta at 300kV 2) This Ceta has 4x better SNR compared to a normal Ceta.

How is the CETA reaching such high resolution? Higher mag to push to better DQE region?

It is really easy, as easy as Krios/Glacios, basically only to put grids into the cassette. 5 grid/hour?🤔We need to put you to the test😂
Seriously, I like the Tundra!🔬🙂👍 If the loading is indeed very easy and quick, put a plunger next to it and plunge-load-plunge-load-... until you get the perfect grid for the bigger microscopes.😍
